I do, though for the first page I started coloring today, I began with the first panel, fully coloring it, then went for particular areas across the page. It's been a while since I colored a comic book page (outside of my single cartoon panels) so I expect to use the "flats" as a good start to lock in the page.

I use Photoshop, but have not done a lot of coloring of comicbook type images. What I have done took a lot of time though.
